GAMBARAN RÖNTGEN DAN HISTOPATOLOGI GIGI MOLAR TIKUS (Rattus norvegicus) YANG DIINDUKSI KARIES GIGI YANG DIBERI PASTA GIGI DAUN BELUNTAS (Pluchea indica)

Abstract:Dental caries is one of the most prevalent oral diseases. Dental caries can be
initiated by many factors such as Streptococcus mutans and sucrose consume.
Pluchea indica leaves contain of anti-inflammation, anti-oxidant, and broad
spectrum of antimicrobial. The purpose of this research was determine the ability
of Pluchea indica extract tooth paste to decrease of Streptococcus mutans growth
causing dental caries by in-vivo examination (rӧ ntgen and histopathology). In this
research, 25 rats were use. This rats was divided into 5 groups. Group 1 was
negative control given by base diet. Group 2 was a positive control without
treatment, given caries diet and induced by Streptococcus mutans in first week of
research. Group 3-5 given caries diet, induced by Streptococcus mutans and
treated by Pluchea indica extract tooth paste for 40%,50% and 60% respectively,
the treatment were given from 8th until 10th weeks of research. At the end of
research, all rats were euthanized by chloroform and necropsied for rӧ ntgen and
histopathology examination. Based on rӧ ntgen result, there was a difference of
contrasting colors and showed the difference among experimental groups, group 4
reviled radiopaque and showed similar to the negative control group. Result of
histopathology examination showed differences in the structure and thickness in
among experimental groups. However, group 5 (60% Pluchea indica leaves
extract tooth paste) that showed similar structure and thickness. This happen
because of Pluchea indica extract paste can reduce amount of bacteria initiated
dental caries. Result of research showed Pluchea indica extract tooth paste can
reduce dental caries.
Intisari:Karies gigi merupakan salah satu penyakit mulut dengan prevalensi tinggi.
Karies gigi dapat disebabkan oleh berbagai faktor seperti pola konsumsi sukrosa
dan Streptococcus mutans pada gigi. Daun Pluchea indica memiliki kandungan
sebagai antiradang, antioksidan, dan antimikroba berspektrum luas. Tujuan
dilakukan penelitian ini adalah untuk mengetahui kemampuan pasta gigi ekstrak
daun Pluchea indica dalam mengurangi pertumbuhan Streptococcus mutans yang
dapat menyebabkan karies dengan uji in vivo (rӧ ntgen dan histopatologi). Uji in
vivo dilakukan dengan membagi tikus menjadi 5 kelompok. Kelompok 1
merupakan kontrol negatif, tikus yang diberi diet biasa. Kelompok 2 merupakan
kontrol positif, tikus diberi diet karies dan diinduksi Streptococcus mutans pada
minggu pertama penelitian. Kelompok 3-5, tikus diberi diet karies, diinduksi
Streptococcus mutans dan diberi perlakuan sikat gigi dengan pasta gigi ekstrak
daun Pluchea indica 40%, 50% dan 60% yang diberikan sekali sehari pada
minggu ke 8-10 penelitian. Pada akhir penelitian, semua tikus di nekropsi untuk
diambil foto rӧ ntgen dan di uji histopatologi dengan pewarnaan Hematoxylin-
Eosin. Hasil rӧ ntgen menunjukkan perbedaan warna kontras antar kelompok,
terutama pada kelompok 4 (pemberian 50% pasta) menunjukkan hasil yang mirip
dengan kontrol negatif. Hasil histopatologi menunjukkan terjadi perbedaan
struktur dan ketebalan gigi pada setiap kelompok, terutama pada kelompok 5
(yang diberi 60% pasta) menunjukkan hasil yang mirip dengan kontrol negatif.
Hal ini dapat terjadi karena pasta gigi ekstrak daun Pluchea indica dapat
mengurangi jumlah bakteri yang dapat menginisiasi karies gigi. Hasil penelitian
menujukkan bahwa pasta gigi ekstrak daun beluntas mampu menghambat karies
